Preparation of 200 mL of 70% (v/v) ethyl alcohol

Introduction

  • Histopathology mein, alcohol solutions ka use tissue ko preserve karne aur fix karne ke liye hota hai.
  • Ethyl alcohol, jo ki commonly histopathology labs mein use hota hai, kaafi important role play karta hai tissue fixation, dehydration, aur staining processes mein.
  • 70% (v/v) alcohol solution ka preparation kaafi zaroori hai kyunki yeh tissue ko fix karne, degradation se bachane aur achhe se penetration dene mein madad karta hai.
  • Yeh procedure batata hai kaise 200 mL 70% (v/v) ethyl alcohol solution prepare kiya jaata hai, jo commercially available ethyl alcohol se banaya jaata hai, jo usually 95% ya 100% concentration mein available hota hai.
  • Is process mein, hum higher concentration wale alcohol ko dilute karte hain taaki hum 70% alcohol solution bana sakein, jo histopathology mein commonly use hota hai.

 

Materials and Requirements

Materials:

  1. Commercial Ethyl Alcohol (100% ya 95%)

    • Yeh histopathology labs mein commonly available hota hai.

    • Usually concentrated form mein (100% ya 95%).

  2. Distilled Water

    • Alcohol ko dilute karne ke liye use hota hai.

  3. Graduated Cylinder ya Pipette

    • Liquid volumes ko accurately measure karne ke liye.

  4. 200 mL Graduated Flask

    • Final alcohol solution prepare aur mix karne ke liye.

  5. Stirring Rod

    • Alcohol aur water ko achhe se mix karne ke liye.

  6. Protective Gloves

    • Chemicals handle karte waqt safety ke liye.

  7. Labeling Material

    • Prepared solution ko label karne ke liye.

Equipment:

  1. Measuring Cylinder (Accurate volume measurement ke liye).

  2. Graduated Flask (200 mL capacity).

  3. Stirring Rod (Uniform mixing ke liye).

  4. Pipettes ya Syringes (Agar precision chahiye ho).

 

Procedure

  1. Alcohol ki Volume Calculate Karna:

    • Dilution formula C1×V1=C2×V2

      • = Initial alcohol concentration (100% ya 95%),

      • = Volume of alcohol jo required hai,

      • = Desired final concentration (70%),

      • = Final volume (200 mL).

    • 100% Alcohol ke liye:

      100 \times V_1 = 70 \times 200 \implies V_1 = 140 \, \text{mL of 100% alcohol}.

      To, aapko 140 mL of 100% ethyl alcohol chahiye.

    • 95% Alcohol ke liye:

      95 \times V_1 = 70 \times 200 \implies V_1 \approx 147.37 \, \text{mL of 95% alcohol}.

      To, aapko 147.37 mL of 95% ethyl alcohol chahiye.

  2. Alcohol Measure Karna:

    • Graduated cylinder ya pipette ka use karke 100% ya 95% alcohol ka required volume measure karein:

      • Agar 100% alcohol use kar rahe hain, to 140 mL measure karein.

      • Agar 95% alcohol use kar rahe hain, to 147.37 mL measure karein.

  3. Distilled Water Add Karna:

    • Distilled water ko add karein taaki total volume 200 mL ho jaye:

      • Agar 100% alcohol use kar rahe hain, to 60 mL of distilled water add karein.

      • Agar 95% alcohol use kar rahe hain, to 52.63 mL of distilled water add karein.

  4. Solution Mix Karna:

    • Alcohol aur distilled water ko 200 mL graduated flask mein daalein.

    • Stirring rod ka use karke mixture ko achhe se mix karein taaki uniform solution ban jaye.

  5. Final Volume Verify Karna:

    • Mix karne ke baad, ensure karein ki final volume exactly 200 mL ho. Agar zaroorat ho, to thoda aur distilled water add kar sakte hain.

  6. Solution Label Karna:

    • Solution ko label karein: “70% Alcohol Solution”, date of preparation, aur alcohol concentration mention karein. Proper labelling is important hai taaki future mein confusion na ho.

  7. Storage:

    • Prepared 70% alcohol solution ko sealed container mein store karein, cool, dry place mein, door from direct sunlight, heat, ya open flames. Ethyl alcohol is highly flammable hota hai, isliye safety precautions zaroori hain.

 

Applications 

  1. Tissue Fixation:

    • 70% alcohol solution tissue fixation mein use hota hai. Yeh tissue ki structure ko preserve karta hai, enzyme degradation ko rokta hai, aur cellular morphology ko maintain karta hai for further histological examination.

  2. Tissue Dehydration:

    • Alcohol in different concentrations mein tissue dehydration ke liye use hota hai. 70% alcohol solution aksar ek series mein use hota hai, jisme gradually alcohol ki concentration increase hoti hai, taaki tissue se water nikaal sakein.

  3. Clearing Agent:

    • Alcohol clearing agent ke roop mein bhi use hota hai. Yeh tissue ko paraffin ya resin mein embedding ke liye prepare karta hai, water ko remove karne mein madad karta hai.

  4. Staining Procedures:

    • Alcohol tissue samples ko staining procedures ke liye prepare karne mein madad karta hai, jaise ki Hematoxylin aur Eosin (H&E) staining.

  5. Tissue Preservation:

    • Alcohol solutions, especially 70%, tissue specimens ko long-term preservation ke liye use hote hain, jisse tissue samples ko store karke unki integrity maintain ki ja sakti hai.
